研究领域
My research in Wuhan Botanical Garden will focus on evolutionary genomics and comparative functional genomics of Solanaceous and Berberidaceous species. For the Solanaceous species, genome duplication, especially the large fragment duplication, will be studied based on the known EST and genomic sequences and marker information from SGN (www.sgn.cornell.edu). Tomato paralogs and corresponding orthologs will be computationally identified and confirmed in the wet lab through mapping and sequencing. Gene expression and function of paralogs and orthologs will be studied through Northern, RT-PCR, and gene transformation. The Epimedium genus in Berberidaceae has about 57 identified species, with 47 species known as YinYangHuo and widely distributed in China. Although Epimedium species are traditional Chinese medicinal plants used for hypogonadism, spermatorrhea, improvement of immune function etc., little is known about these plant species, especially at the molecular level. The genetics studies are proposed for Epimediums, including molecular systematics of Epimedium species, natural genetic diversity evaluation of major species used for medicinal purpose, heredity of biosynthesis of phytochemicals, conservation of Epimediums, cultivar selection from wild species and molecular breeding. cDNA libraries will be constructed using young and mature leaves, flower, and roots tissues. EST sequences will be analyzed for genome characterization, gene expression profile, and identification of genes related to secondary metabolism. Ortholgous genes related to biosynthesis of phytochemicals will be explored in the close related Mahonia and Berberis species, that are also traditional Chinese medicinal plants, and compared with other model species. Bioinformatics component (e.g. databases, websites, and sequence analysis) will be developed for storing, management, and analysis of sequences, annotation results, and natural diversity.
