臧敬五教授的研究是立足于从临床问题中提炼出关键免疫学科学问题,用最新的科学手段在分子和细胞水平上进行研究,并致力于成果的临床应用。
一、通过对髓鞘抗原自身反应T细胞的研究,揭示了其在体内激活的可能机理及在多发性硬化炎症和中枢神经系统脱髓鞘病理过程中的关键作用。
二、首创用灭活髓鞘抗原反应T细胞治疗多发性硬化(T细胞疫苗)。初步阐明了其体内免疫调节机理,并正在探索T细胞疫苗在多发性硬化和类风湿关节炎免疫治疗中的临床应用前景,研制T细胞受体多肽治疗性疫苗。
Our most recent research interest is briefly described as follows
Role of regulatory T cells in autoimmune conditions. Our research interest focuses on the basic mechanisms involved in the induction and conversion of CD4+CD25+ Treg by Copolymer-I (PNAS, 102: 6449-6454, 2005), gamma-interferon (JCI, In press, 2006) and T cell vaccination (PNAS, 103: 5024-5029, 2006). Using a humanized mouse model, our research expands to the role of thymus in the development and differentiation of CD4+CD25+ regulatory T cells. The other aspect of our research in this area is related to elucidation of signaling events involved and cytokines required for the function of CD4+CD25+ regulatory T cells in humans and rodents.
Role of autoreactive T cells in multiple sclerosis and rheumatoid arthritis and the development of novel treatment strategies to target at autoreactive T cells. Recently completed studies include: T cell receptor V gene analysis (Genes and Immunity, 6: 248-261, 2005), role of osteopontin as a key pro-inflammatory cytokine (JCI, 115: 1060-1067, 2005), involvement of MAPKK pathways (Journal of Immunology, 173: 7299-7307, 2004), functional properties of CD8+ autoreactive T cells (Journal of Immunology, 172: 5120-127, 2004) and stem cells (Brain, 127: 996-1008, 2004). Several lines of research are on-going to develop novel treatment strategies for MS and RA, which include T cell vaccination (Arthritis & Rheumatism and PNAS, 2006) and the use of neural stem cells or anti-inflammatory small molecules from Chinese herbal medicine.
