Brief Introduction to the ongoing research projects
1. Developing the techniques which have potential use in DNA CHIP preparations such as:
*Temperature dependent cross-linking chemistry was proposed and confirmed its application in immobilizing DNA fragments or oligonucleotides onto the surface of glass slide. This technoque might be better then the reported methods in cross-linking speed and efficiencies.
* The porous plastic was used as substrates for DNA chip preparations. The advantages of porous substrates are the surface area of each sample spot is much larger then the flat substrate, so that much more probes can be immobilized on each spot. As more samples can be immobilized on each spot, the hybridization speed is much fast (30 minutes are enough), and the spot signal is much stranger then flat substrate (several decade stronger). Porous plastic should be better then porous glass as (1) it is easier to be processed; (2) plastic is cheaper then glass; and (3) the structure can be definitely controlled.

2. Screening the genes that related to the brain development using cDNA microarray. Total mRNAs were isolated from rat brain and reverse transcribed into cDNAs. The cDNAs were cloned into vectors to make the library. The clones of the library were uniformed to delete all possible repeated clones. About 10,000 clones were obtained and amplified with PCR reactions. The amplified DNA fragments were spotted onto the slides to prepare the DNA chips and then the gene expression patterns of rat brains at different development stages were analyzed to characterize the gene related to brain development.
